Mutation Detection - Variance Analysis Services

Currently the Core Lab offers Mutation Detection or Variance Analysis Services using PCR amplification and DNA Sequencing to investigate genomic DNA regions or genes of interest.
If you are looking for other than PCR/sequencing base approach for your variance detection project please contact us directly.

We have substantial experience designing and optimizing PCR primers and using dedicated analysis software for mutation detection. We also make use of lab automation for high throughput projects.
In summary we offer a complete service at a great value and fast turnaround time so that investigators and their lab staff can focus on more productive aspects of their research.
You bring us the genomic DNA extraction and we’ll take care of the rest!

Mutation Detection Project Workflow

  • Project Desgin: decide on sequencing strategy, design and order primers.
  • PCR Optimization: use internal control samples to test and optimize PCR conditions. PCR products are sequenced for validation. Re-design primers if necessary.
  • PCR sample amplification: generate PCR for samples in 96 or 384-well plates.
  • DNA Sequencing: sequence PCR products using forward and reverse primers.
  • Mutation Analysis: Contig, align and compare sequence data against a chosen standard reference. Generate mutation reports.
  • Finishing: Validate mutations.

Clarifications:

  • Primers are order either from the on campus Oligo Core or from IDT.
  • PCR Optimization plates: We can optimize up to 15 primer sets per plate. The lower price is for testing up to 7 primers, the higher price is from 8 up to 15 primer pairs. About 80% of the primers tested work the first time.
    Note that if we have already develop the primers for you, there is not optimization charge.
  • The cost of PCR includes the PCR reaction itself, agarose gel check and PCR clean-up. There is a minimum charge of $50 regardless of sample number.
  • Cost of Sequencing: see prices and volume discounts for regular sequencing services.
  • Charges for Analysis are rounded up to the next hour.

Cost Estimate Example:
Supose that an investigator wants to sequence all the 12 exons of a gene of interest for a total of 30 samples. Let's assume that 10 primer pairs are necessary to cover all the exons and 3 PCR optimation plates were necessary to get all the primers tuned up:
Primers: ~$150
PCR Optimization: $300
PCR of samples: $480 (samples + controls)
Sequencing: $2,240 (640 sequences- including controls - @ $3.5 each)
Analysis: $60 (3 hr)
Total Estimate for project: $3,230
