The Core Facility is equipped with a ABI 3730XL high throughput Capillary Electrophoresis instrument dedicated exclusively to DNA Sequencing.
Turnaround
We strive to provide you with 24hr turnaround. If samples are submitted in the morning they will be sequenced by the next day and posted online before noon. You will receive an email from the system when samples are posted.
Note: since we process samples on batches of 96, the very last orders submitted before the cutoff time might need to be put on hold until we have more samples from other users to fill a 96-well plate.
Providing we receive enough samples in the afternoon, we will process another batch later in the day and have the results posted in 24hr.
Sample Submission
- Use the correct containers to submit samples. Please use only the tubes and plates below. We have them available for purchase at the Core facility. See Changes Dec '07.

Tubes with

- Every tube must be labeled with Tough-Tags. Clearly label the tubes numericaly (1,2, 3, etc...) followed by the sample name.
- Keep caps on a strip. The caps are pre-split. You can pipette into the tubes after capping them.
- Add a piece of tape with the last 5 digits of the order ID on top of the tubes!

- Label the plate with the order name.
- Starting A01, fill the plate by column
A1, B1, C1....H1, A2, B2 and so forth.
- Please, no empty wells in between samples.
- Leave position H12 empty for our control.
- Cover the plate with Sealing Film.

- Add the right amount of DNA, primer to tubes or plates according to the table below. Please do not submit less than 10 ul total volume. Primer must be added by the user.

Important Notes:
- The facility provides the following Universal Primers: M13F, M13R, SP6, T7, T7Terminal and T3. See primer sequences. It is the user responsibility to add the primer to her/his samples.
- All DNA should be check for concentration and purity before they are submitted to the Core. Both by OD as well as agarose gel. We will not reprocess for free samples that have obvious problems with concentration and/or purity.
- We recommend DNA templates, both PCR and plasmids, to be purified using prep kits from well established brands such as Qiagen, Invitrogen, Millipore, Amershan, etc. Excessive salt, alcohol, protein and RNA will compromise the sequencing reaction. Left over primer and dNTPs from PCR reactions will also interfere with DNA sequencing.

We encourage every user to have his/her own log in account. Please avoid seting up a "common" account for the whole lab.
- Drop off your samples at one of the assigned locations.
Off-campus users please FedEx your samples to our lab. Samples should be fine shipped at RT.
- You will receive an email from our system when the sequences are ready for download.
